The protocols listed below (in pdf format) are
standard protocols that we have routinely used but of course are not by any
means necessarily the ‘best’ way to prepare the material in question, as
methodologies are undoubtedly constantly improving and different
investigators have their own preferences with regards to reagents and so
on. Having said this, we would
ask that investigators submitting their DNA ready for ES cell electroporation at least adhere to the basic principal
that the DNA should be thoroughly repurified after endonuclease-mediated linearization. This will help to ensure an optimal
ES cell electroporation for homologous recombinant
clone production.
