Mouse Embryonic Stem Cell and
Transgenesis Core Laboratories
Policies
Investigator's Responsibilities
For DNA injection: In general terms, the investigator must
prepare the DNA to be injected and assume responsibility for screening and
care of the weaned animals. Specifically, in order to provide you with this
service, it is requested that you:
-
Contact Ms. Gabriela Pacholczyk (721-1226) to discuss the schedule of
injections.
-
Provide the core with a high quality DNA preparation of your construct
with documentation of its purity. A collection of a 0.5-ml tubes
appropriately labeled and containing 10 mg or more of DNA.
-
Fill out and sign a service request form, including evidence of approval
of the CAURE committee for the project.
-
Provide analysis of tail biopsies within 10 days so that positive animals
can be identified in a timely manner and cage costs minimized. After 10
days, the investigator will be charged for cage costs at a rate of $1.00
per day per cage as long as the animals remain in the care of the core
facility.
-
Undertake care of the transgenic pups submitted to the
project scientist. These mice can no longer
reenter the barrier colony. It is the responsibility of the individual
investigator to arrange for animal housing.
For ES Cell Injection: The ES
Cell core is responsible for maintaining the recombinant ES clone under
optimal conditions that would inhibit their differentiation. It is
recommended that you:
-
Contact Ms. Suzanne Morris (721-1226) to arrange for electroporation and
discuss the schedule of injection.
-
Fill out and sign a service request form, including evidence of approval
of the CAURE committee for the project. Unless you have been involved with
chimeric mouse work, this will require a new submission to the committee.
-
Upon notification of a prearranged schedule, provide the core personnel with a
freshly harvested sample of linearized targeting construct at least one
week in advance of the electroporation date.
-
Provide lyses buffer for DNA harvesting 10-12 days after the
electroporation day.
For embryo and sperm freezing:
The investigator should contact Ms. Gabriela Pacholczyk (721-1226) to
discuss the plans for the service. These would include the availability of
breeding pairs, the possibility of admitting the animals into the barrier
facility, the number of mouse lines involved, and the service fees.
Policies
Committee Approval: It is an institutional requirement that
every investigator demonstrate an updated approval by the Institutional
Animal Care and Use committee (CAURE) prior to using the Transgenic & ES
Cell Core Laboratories.
Scheduling: It is our policy that no injection or
electroporation date will be assigned to any DNA construct until it is
submitted along with a completed service request form and the required
documents attached.
Confidentiality: All information provided by the investigator
concerning the research project is treated with the utmost confidentiality.
No subject related material will be released to or shared with anyone
without a written consent of the principal investigator.
Support Letter: For those investigators preparing to submit
grant proposals to funding agencies, we will be happy to provide you with a
letter stating our readiness to make transgenic and/or chimeric mice for
your project.
Acknowledgments: We request that any scientific publications
involving these transgenic animals acknowledge the service of Medical
College of Georgia Transgenic & ES Cell Core Laboratories in the Materials and
Methods section.
