Genomics Core Laboratory
DNA Sequencing

The Genomics Core Laboratory (GCL) at the Medical College of Georgia (MCG) offers automated fluorescence DNA sequencing using a state-of-the-art ABI 3730 XL 96-capillary sequencer. This is an ultra-high through-put instrument capable of read lengths over 850bp with accuracies approaching 100%. Our default chemistry is ABI’s Big Dye Terminator 3.1. This allows for greater sensitivity and longer reads. If you wish to use another chemistry, please contact the facility.

We request that samples be submitted as follows:

  • Plasmid DNA- at least 300ng in no more than 10μl of molecular biology grade water. No TE please, this might inhibit the reaction.

  • PCR Product- at least 40ng in no more than 10μl of molecular biology grade water. Be sure that any residual primer from the PCR reaction has been removed. No TE please, this might inhibit the reaction.

  • BAC DNA- at least 1mg of DNA in no more than 15μl of molecular biology grade water. No TE please, this might inhibit the reaction.

Our turn around time is, on average, 72 hours. Any samples received after 3:00pm will not be processed until the following day. Pricing is currently $15 per reaction. This includes all reagents, reaction purification, array run and data.

Results will be E-mailed or placed on a server for the customer to pick up as soon as they are ready. The text file of the sequence can be viewed in any text program. To view the electropherogram, you will need a viewing program for this type of file. These can be found at several locations including the following:

Edit View for Mac:

http://www.appliedbiosystems.com/support/software/dnaseq/installs.cfm

or Finch Trace View for Windows or Mac OS X and Redhat/SuSE Linux:

http://www.geospiza.com/finchtv/

For PC, use Chromas, this can be downloaded from:

http://www.technelysium.com.au/chromas14x.html

This will allow you to view as well as edit and print these files.

All samples submitted must be accompanied by a sample submission form. This form must be completely filled out so that sample processing will not be delayed.

Please specify if you wish for your samples to be returned. They will be held for seven business days and then destroyed. On-campus users may pick them up in the core. Return postage fees will apply for off-campus users.
